Rare endemic plants of the mountainous Crimea <i>Crepis purpurea </i>(Willd.) M. Bieb.) and <i>Scrophularia exilis</i> Popl. preservation under <i>in vitro</i> genebank conditions

The article presents for the first time results of a study preservation features explants 2 rare endemic species under conditions in vitro genebank 12 months. Microshoots Crepis purpurea (Willd.) M. Bieb (Asteraceae) and Scrophularia exilis Popl. (Scrophulariaceae), cultured were included research. Microshoots’ segments 0,5-1,0 cm long used as preservation. placed on an agarized culture medium with recipe ¼ MS supplemented growth inhibitors: 0,2 g/l CCC chlorocholinchloride (BASF, Germany) 60 g / l sucrose (Panreac, Spain). As control, was used, without retardants. Culture vessels refrigerators light intensity 1.25-3.75 mcM m -2 s -1 temperature 4, 6, 8, 10 12,0°C. Plant material evaluated after 6 months cultivation using qualitative quantitative characteristics explants. It found that viability reduction kinetics during deposition genebank, along low illumination use nutrient inhibitors, facilitated by exposure to 4,0-6,0°C. Conservated studied genotypes transferring standard cuture morphogenetic potential ability regenerate microshoots microrosettes remained.
